N-terminal 6His-tagged, recombinant, TAO2. TAO2 is a mitogen-activated protein kinase kinase kinase (MAP3K) that doubly phosphorylates and activates the MAP kinase kinases (MAP2Ks) MEK3 and MEK6.

10 µg of enzyme in 100 µL of 50 mM Tris/HCl pH7.5, 300 mM NaCl, 0.1 mM EGTA, 0.03% Brij 35, 270 mM sucrose, 1 mM benzamidine, 0.2 mM PMSF, 0.1% 2-mercaptoethanol. Frozen solution.

Quality
Routinely evaluated by phosphorylation of 0.8 mg/ml myelin basic protein (MBP). Purity was assessed by SDS-PAGE and Coomassie blue staining using 3 µg of TAO2.
Storage and Stability
Stable for 6 months at -70°C from date of shipment. For maximum recovery of product, centrifuge original vial prior to removing the cap.
Rapidly thaw the vial under cold water and immediately place on ice. Aliquot unused material into pre-chilled microcentrifuge tubes and immediately snap-freeze the vials in liquid nitrogen prior to re-storage at -70°C.

Specific Activity: where one unit of TOA2 activity is defined as 1 nmol phosphate incorporated into 0.8 mg/mL myelin basic per minute at 30°C with a final ATP concentration of 100 µM.
